IscU1/2 Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects IscU1 and IscU2 in human samples as well as IscU in mouse and rat samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). IscU1/2 (D-6) antibody is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. IscU proteins play a crucial role in the assembly of iron-sulfur (Fe-S) clusters, which are essential cofactors involved in electron transfer in oxidative phosphorylation and enzymatic reactions in key metabolic pathways. IscU proteins are primarily localized in the mitochondria, where Fe-S cluster assembly occurs through interaction with IscS, a cysteine desulfurase that provides inorganic sulfur. Deletion of IscU1 and IscU2 can lead to iron accumulation in mitochondria, loss of aconitase activity, and increased oxidative damage in cells lacking cytosolic copper/zinc superoxide dismutase. Alternative splicing of IscU pre-mRNA results in IscU1, which localizes to the cytosol, and IscU2, which is found in the mitochondria, highlighting the importance of subcellular localization in Fe-S cluster assembly regulation.
