iCRT 14 is a potent inhibitor of beta-catenin-responsive transcription (CRT) (IC50 = 40.3 nM in assays of Wnt pathway activity). The Wnt pathway is a cell signaling system involved in regulating cell fate, proliferation, migration, and organogenesis during embryonic development. iCRT 14 influences the interaction between β-catenin and TCF4. β-catenin is a key mediator in Wnt signaling; upon Wnt activation, stabilized β-catenin translocates to the nucleus, where it forms a complex with TCF and other co-factors to activate the transcription of Wnt target genes. By blocking the β-catenin/TCF interaction, iCRT 14 effectively inhibits the transcriptional activation of Wnt target genes, thereby modulating the pathway′s activity.
