Hello, I want to use this antibody for immunostaining GLUT-4 in nonpermeabilized cells to selectively stain the surface GLUT-4. Should this antibody work for my experiment? I am curious whether this antibody recognizes extracellular epitope of GLUT-4.

Thank you for your question. The exact epitope recognized by this mouse monoclonal antibody has not yet been determined. We have not tested this antibody in non-permeablized cells. You may want to check some of the product citations using this antibody to see if others have previously published in the same application.

Thank you for your question. We do offer this mouse monoclonal, Glut4 Antibody (IF8): sc-53566 directly conjugated to FITC or fluoroscein, the catalog number is sc-53566 FITC. We have not tested this antibody for flow cytometry, however we are aware of a publication citing use of the antibody for this application (on PubMed, PMID: 23074218). Based on this publication, we would recommend the antibody for flow cytometry.

Rated 1 out of 5 by from Low Signal and High Background I used the Glut4 Antibody (IF8): sc-53566 HRP on rat skeletal muscle (soleus and EDL) and white adipose tissue for Western blotting. The signal was very faint and there was a lot of background. I loaded 50-75 ug protein, used a 1:200 antibody dilution in 5% milk, and blocked in 5% milk.

Rated 5 out of 5 by from relatively clean, strong signal This antibody successfully detected GLUT4 in mouse brown adipose tissue lysates (20 ug/lane). Lysates were not boiled, but were reduced. I used this antibody at a 1:200 overnight @4C. I imaged with Li-COR Odyssey system which was able to detect signal without issue.
