AChE Antibody (A-11) is a mouse monoclonal IgG2a kappa light chain antibody that detects acetylcholinesterase (AChE) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). AChE monoclonal antibody (A-11) is raised against amino acids 481-614 of AChE of human origin. Anti-AChE antibody (A-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Acetylcholinesterase plays a crucial role in the nervous system by hydrolyzing acetylcholine at synaptic junctions, thereby terminating synaptic transmission and ensuring proper neuronal signaling. AChE exists in three forms due to alternative mRNA splicing: the T form, which is soluble and found in synapses; the H form, which is globular and located on the outer surfaces of cell membranes; and the R form, which is not functionally characterized. The H form′s subunits are glycosylphosphatidylinositol-anchored to cell membranes, while the T form′s subunits are tethered to basal lamina components via a collagen tail. Loss of AChE activity in cholinergic and noncholinergic neurons is associated with Alzheimer′s disease, where increased AChE activity is observed around amyloid plaques, potentially linked to disturbances in calcium homeostasis and the activation of L-type voltage-dependent calcium channels.
