
PRAM-1 Antibody (A-5) is a mouse monoclonal IgG1 antibody that detects PRAM-1 in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PRAM-1, or PML-RAR-α target gene encoding an adaptor molecule 1, plays a crucial role in normal human myelopoiesis, acting as an adaptor protein that facilitates signaling pathways essential for hematopoietic cell development. PRAM-1 expression is significantly modulated during this process, and PRAM-1 expression is notably hindered by the presence of the PML-RAR-α fusion protein, which is implicated in acute promyelocytic leukemia. The 718 amino acid structure of PRAM-1 features eight N-terminal proline-rich repeats and several proline residues that form type I and type II SH3 recognition motifs, which are vital for protein-protein interactions. This structural characteristic allows PRAM-1 to interact with specific signaling molecules, thereby influencing cellular responses and maintaining normal hematopoietic function. PRAM-1 is predominantly expressed in hematopoietic tissues and lung, highlighting PRAM-1′s significance in both normal physiology and disease states.
