Nfix encodes the nuclear factor I X-type (NF-1X), a sequence-specific DNA-binding transcription factor that recognizes NFI consensus elements to regulate promoter and enhancer activity during development. In mouse, NF-1X contributes to lineage specification and maturation programs in the nervous system and skeletal muscle, influencing cell-cycle exit, differentiation, and extracellular matrix–associated gene expression. NF-1 family factors interface with chromatin remodeling and transcriptional co-regulator networks to coordinate developmental timing and tissue architecture. Dysregulated NFIX function has been linked to developmental syndromes and altered growth and differentiation phenotypes, making Nfix a useful node for studying gene regulatory circuits relevant to neurodevelopment and myogenesis.
NF-1X CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Nfix gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Nfix together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Nfix open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish NF-1X protein expression.
This CRISPR knockout system enables efficient generation of Nfix-deficient cell models for investigation of NF-1X signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
