myocilin Antibody (F-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects myocilin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-myocilin antibody (F-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Myocilin is an extracellular protein predominantly expressed in the eye, particularly in the retina, trabecular meshwork, and ciliary body, where myocilin plays a crucial role in maintaining intraocular pressure (IOP). Myocilin′s ability to form homomultimers in vivo and interaction with components of the extracellular matrix, such as the Hep II domain of fibronectin, highlight myocilin′s importance in structural integrity and cellular signaling within the ocular environment. Additionally, myocilin interacts with myosin regulatory light chain, a key component of the myosin motor protein complex, suggesting myocilin′s involvement in the actomyosin system, which is essential for the proper functioning of the trabecular meshwork. Disruptions in this system can lead to elevated IOP and contribute to the development of glaucoma, a leading cause of blindness worldwide. Myocilin is encoded by the MYOC gene, also known as TIGR, located at the GLC1A locus on chromosome 1q24.3, and mutations in myocilin are specifically associated with primary open-angle glaucoma (POAG), a progressive condition characterized by the loss of retinal ganglion cells.

The MYOC gene is highly homologous between mouse and human, I wonder this antibody can differentiate them, especially for transgenic mouse with human MYOC, thank you.

Thank you for your question. This antibody can not distinguish between mouse and human MYOC or myocilin proteins. The antibody is recommended for detection of mouse, rat and human myocilin proteins.

Do you have a reference related to development of this antibody? Additionally, could you refer me to an example/reference for use of this antibody in ELISA? Thank you.

Thank you for your question. This antibody is a Santa Cruz product and was developed in house. We do not have a published citation in ELISA that we are aware of, but here are two citations using it in Western blot (PMID: 24691439, 21821918).
