Ceruloplasmin Antibody (3B11) is a mouse monoclonal IgG1 antibody that detects ceruloplasmin in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Ceruloplasmin is a vital blue plasma glycoprotein synthesized in hepatocytes, playing a crucial role in copper transport throughout the body. Ceruloplasmin, also known as ferroxidase, is composed of three homologous domains resulting from an intragenic triplication, and exists in two splice variants, CP-1 and CP-2, which exhibit differential expression in specific tissues. Ceruloplasmin mRNA is notable in human liver, macrophages, and lymphocytes, highlighting widespread biological significance. Ceruloplasmin binds six or seven cupric ions per molecule and is essential for the peroxidation of Fe(II) transferrin to form Fe(III) transferrin, a critical step in iron metabolism. The structural integrity of ceruloplasmin is crucial for function; while the proteolytically degraded short form retains some ferroxidase activity, only the intact long form can effectively catalyze iron loading into ferritin. Ceruloplasmin induces low-density lipoprotein oxidation in vitro, a process that relies on the presence of a single, chelatable copper atom. A glycosyl phosphatidylinositol (GPI)-anchored variant of ceruloplasmin is expressed by Sertoli cells, suggesting potential significance in reproductive biology. With multifaceted roles in copper and iron metabolism, ceruloplasmin is critical for maintaining homeostasis in the human body, making anti-ceruloplasmin antibody (3B11) an invaluable tool for researchers studying these essential processes.
