CD3-ε Antibody (48-2B) is a hamster monoclonal IgG antibody that detects CD3-ε in mouse and rat samples through applications such as immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). CD3-ε plays a crucial role in the T cell receptor (TCR) complex, which is essential for T cell activation and immune response. Located on the surface of T cells, CD3-ε is one of the five invariant polypeptide chains that form the CD3 complex, which is vital for transmitting signals from the TCR upon antigen recognition. The presence of CD3-ε, along with its associated chains, facilitates the clustering of the TCR and the subsequent activation of intracellular signaling pathways that lead to T cell proliferation and differentiation. Importantly, CD3-ε contains an immunoreceptor tyrosine-based activation motif (ITAM) that is critical for the recruitment of signaling molecules, such as ZAP-70, which further propagate the activation signal. Can't make this antibody work on mouse FFPE spleen tissue. Have tried various pre-treatment (EDTA, citrate) but no signal so far using 1/30 dilution and 1hour incubation.Can someone recommand a protocol that works? Thank you.

Can CD3-ε (48-2B): sc-1174 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD3-ε (48-2B): sc-1174 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
