Amphiphysin II Antibody (99D) is a mouse monoclonal IgG2b kappa light chain antibody that detects Amphiphysin II across mouse, rat, human, and avian species through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-Amphiphysin II antibody (99D) is available in a non-conjugated form for versatile experimental use. Amphiphysin II is a brain-enriched protein playing a significant role in synaptic vesicle recycling, essential for neurotransmission. Amphiphysin II (99D) monoclonal antibody recognizes a protein that functions as a dimer and features a membrane-bending BAR domain, a central Clathrin and adaptor binding domain, and a C-terminal SH3 domain. The interaction between Amphiphysin II and the GTPase Dynamin, mediated through the SH3 domain, enables synaptic vesicle recycling in neurons by facilitating Dynamin binding to Clathrin-coated pits, leading to vesicle budding. A splice variant of Amphiphysin II, lacking Clathrin or adaptor interactions, predominantly expresses in muscle tissue and plays a crucial role in T tubule network formation and stabilization, demonstrating diverse functional roles in cellular dynamics.

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. We recommend following our recommended protocol, provided below, https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining Alternatively, consider using our alternative antibody, Amphiphysin II (2F11): sc-23918, which we do offer directly conjugated to HRP and a variety of fluorochromes, if you wish to omit using any secondary detection reagent and potential non-specific binding.
