    Overview

    ACTA1 encodes skeletal muscle alpha-actin, a core component of thin filaments that polymerizes into F-actin to support sarcomere assembly, myofibrillar contractility, and cytoskeletal architecture. Through interactions with tropomyosin, troponin, and actin-binding proteins, ACTA1 contributes to regulated actomyosin dynamics, mechanotransduction, and force transmission in striated muscle. Perturbation of ACTA1 function disrupts sarcomere organization and muscle fiber integrity and is associated with inherited skeletal myopathies, including nemaline myopathy and other congenital muscle disorders. As a highly expressed structural gene in muscle, ACTA1 is also used as a marker in studies of myogenesis, muscle differentiation, and cytoskeletal remodeling.

    ACTA1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ACTA1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ACTA1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ACTA1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ACTA1 protein expression.

    This CRISPR knockout system enables efficient generation of ACTA1-deficient cell models for investigation of ACTA1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
