CAB39L Activators are a class of chemical compounds that enhance the functional activity of CAB39L through indirect mechanisms involving the activation of the AMPK pathway or related upstream regulators. Forskolin, by increasing intracellular cAMP levels, leads to the activation of PKA, which can phosphorylate proteins that regulate the AMPK pathway, of which CAB39L is a critical component. Metformin, Phenformin, Berberine, and Salicylate, as AMPK activators, enhance the activity of CAB39L by increasing the demand for its scaffolding function in the activation process of AMPK. The role of CAB39L in stabilizing the active conformation of AMPK is crucial, and therefore any compound that can activate AMPK, like AICAR and A769662, serves to enhance CAB39L activity.

Resveratrol activates SIRT1, which in turn deacetylates and activates LKB1, a kinase upstream of AMPK. This activation cascade results in the activation of AMPK and an indirect increase in CAB39L functionality due to its association with the AMPK complex. Similarly, D4476 as a CK1 inhibitor, and C2 Ceramide as an activator of PP2A, modulate the activity of LKB1, which is an essential kinase for AMPK activation. Through these actions, they promote the activation of CAB39L. PT1 acts as an allosteric activator of AMPK, enhancing the activity ofCAB39L through its role in the LKB1-AMPK complex. Lastly, Quercetin, through its ability to activate AMPK, supports the functional activity of CAB39L by necessitating its stabilizing presence within the active AMPK complex.
