PIPPIN Antibody (473.1) is a mouse monoclonal IgG1 antibody that detects PIPPIN in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PIPPIN, also known as CSDC2 (cold shock domain containing C2, RNA binding), is a Y-box protein that plays a crucial role in gene expression regulation, particularly in brain tissue where PIPPIN is predominantly expressed. PIPPIN localizes in both nucleus and cytoplasm, containing two RNA-binding motifs (PIP1 and PIP2) along with a cold shock domain, which are essential for PIPPIN′s function as a nucleic acid binding regulatory factor. PIPPIN binds to the 3′-untranslated regions of mRNAs encoding Histone H1 and Histone H3.3, a process vital for proper brain maturation and development. PIPPIN′s interaction with these mRNAs requires coordinated action of all domains, highlighting the structural integrity′s importance. Additionally, PIPPIN undergoes sumoylation in a thyroid hormone-dependent manner, indicating that post-translational modifications may influence regulatory functions in response to extracellular signals, thereby modulating protein synthesis and cellular responses.

Can sc-101108: PIPPIN (473.1) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: DefinitelyNotMatt
Thank you for your question. Yes, sc-101108: PIPPIN (473.1) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
