galectin-3 Antibody (F-8) is a mouse monoclonal IgG2b kappa light chain antibody designed to target galectin-3, also known by its alternate names Mac-2, hMac-2 (human Mac-2), GALBP (galactose-binding protein), CBP35 (carbohydrate-binding protein 35), and LGALS3 (lectin, galactoside-binding, soluble, 3). galectin-3 monoclonal antibody (F-8) recognizes an epitope between amino acids 2-25 at the N-terminus of mouse galectin-3 and is validated for detecting galectin-3 in mouse, rat, and human samples using western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Galectin-3 is a key member of the galectin family of soluble β-galactoside-binding lectins, characterized by a single carbohydrate recognition domain (CRD) that binds to galactose-containing glycoconjugates. This structural feature is essential for galectin-3′s role in mediating critical biological processes such as cell-to-cell adhesion and interactions with the extracellular matrix (ECM). Galectin-3 exists both intracellularly and extracellularly, contributing to diverse functions. Intracellularly, galectin-3 participates in signaling pathways, pre-mRNA splicing, and apoptosis, while extracellularly, galectin-3 facilitates cell adhesion, migration, and interactions with the ECM. These locations enable galectin-3 to regulate processes like tissue development, immune responses, and cellular communication. Additionally, galectin-3 plays a significant role in tumor progression and metastasis by promoting cancer cell proliferation, resisting apoptosis, and enhancing angiogenesis. Galectin-3 interacts with various proteins, including β1 integrins, which are crucial for cell adhesion and migration, the epidermal growth factor receptor (EGFR) involved in cell proliferation and survival, and members of the Bcl-2 family that regulate apoptosis. Galectin-3 is expressed in a wide range of cell types, including colonic and intestinal epithelial cells, inflammatory macrophages, papillary and follicular carcinomas, neoplastic astrocytes, and certain B and T lymphocytes. Elevated levels of galectin-3 are associated with enhanced cancer progression, thyroid malignancies, and Crohn′s disease, making galectin-3 a valuable diagnostic marker for conditions such as Hurthle cell adenomas and carcinomas.
