Selenoprotein O Activators

Selenoprotein O Activators are a collection of chemical compounds that directly or indirectly enhance the functional activity of Selenoprotein O via modulation of selenium bioavailability and cellular redox conditions. Selenocysteine, the pivotal amino acid in Selenoprotein O, is supplied by compounds such as Selenocysteine and Selenomethionine, which provide selenium in a bioavailable form for protein synthesis. The latter, being a dietary selenium source, can be non-specifically incorporated in place of methionine, thus indirectly boosting Selenoprotein O activity. Inorganic selenium donors like Sodium selenite and Sodium selenate are metabolized within cells, contributing to selenocysteine synthesis, while Diphenyl diselenide and Ebselen enhance the cellular selenium pool, with Ebselen mimicking glutathione peroxidase activity and bolstering the antioxidant function of Selenoprotein O.

Additionally, Methylseleninic acid, through its conversion to methylselenol, supports selenoprotein synthesis, and compounds like Phenylarsine oxide, at sub-inhibitory concentrations, can increase the oxidative folding capacity within the endoplasmic reticulum, thereby facilitating correct folding and activity of Selenoprotein O. Tert-butyl hydroperoxide, by inducing oxidative stress, may paradoxically upregulate antioxidant selenoproteins, thus enhancing Selenoprotein O's function. Hydrogen selenide is utilized in the synthesis of selenophosphate, a donor for selenocysteine-tRNA[Ser]Sec, essential for the synthesis and activity of Selenoprotein O. β-Mercaptoethanol contributes to a reducing environment that maintains cysteine residues in their reduced state, aiding in the proper folding of Selenoprotein O. Lastly, Nifuroxazide, by inhibiting STAT transcription factors, could indirectly influence the functional activity of Selenoprotein O by modifying the redox balance and protein folding dynamics within the cell.
