Chemical activators of cyclic nucleotide gated channel subunit beta 1 can facilitate the protein's function by directly or indirectly increasing the levels of cyclic nucleotides within the cellular environment, leading to the channel's activation. Cyclic AMP and cyclic GMP can activate cyclic nucleotide gated channel subunit beta 1 through direct binding. These cyclic nucleotides bind to the regulatory domains of the channel, inducing a conformational change that transitions the channel from a closed to an open state, allowing ions to flow through. The flow of ions through the channel generates electrical signals that are crucial for various cellular processes.

Several chemicals function by enhancing the intracellular concentrations of cyclic AMP or cyclic GMP, thus leading to the activation of cyclic nucleotide gated channel subunit beta 1. For instance, Forskolin activates adenylyl cyclase, which catalyzes the conversion of ATP to cAMP; this increase in cAMP can then activate the protein. Similarly, Zaprinast, Sildenafil, Tadalafil, Vardenafil, and Anagrelide all act by inhibiting different types of phosphodiesterases (PDEs), enzymes responsible for the breakdown of cyclic nucleotides. Their inhibition leads to elevated levels of cGMP or cAMP, thereby promoting the activation of cyclic nucleotide gated channel subunit beta 1. IBMX, which inhibits a broad range of PDEs, prevents the degradation of both cAMP and cGMP, contributing to the activation process. Vinpocetine and Papaverine also block PDEs, leading to increased levels of the cyclic nucleotides and activation of the channel. Rolipram specifically inhibits PDE4, which preferentially hydrolyzes cAMP; its inhibition results in higher cAMP levels that can activate cyclic nucleotide gated channel subunit beta 1. Through these mechanisms, the selected chemicals elevate the concentration of cyclic nucleotides in the cell, which in turn can activate the cyclic nucleotide gated channel subunit beta 1, allowing it to carry out its function in cellular signaling.
