p53 Antibody (2B2.71) is a mouse monoclonal antibody developed as an IgG2b isotype and raised against amino acids 370-378 of the p53 protein of mouse origin, is a highly effective antibody for detecting a conserved, denaturation-resistant epitope in the p53 protein from mouse, rat, and human species. p53 monoclonal antibody (2B2.71) is versatile and works across multiple applications, including western blotting, immunoprecipitation, immunofluorescence, and flow cytometry. Anti-p53 antibody (2B2.71) is available conjugated with either phycoerythrin or fluorescein isothiocyanate, making it ideal for immunofluorescence, immunohistochemistry on paraffin-embedded tissues, and flow cytometry. The p53 protein is critical in cancer prevention and maintaining genomic stability. Encoded by the TP53 gene on chromosome 17p13.1, p53 functions as a tumor suppressor, regulating cell cycle arrest, DNA repair, apoptosis, and senescence. Structural domains of p53, including the Transactivation Domain (TAD), DNA-Binding Domain (DBD), Oligomerization Domain (OD), and Regulatory Domain, are vital for these processes, with post-translational modifications further controlling its activity during cellular stress. Mutations in the TP53 gene are among the most frequent in human cancers, occurring in over half of tumors. Such mutations compromise p53′s ability to regulate cell growth, leading to uncontrolled proliferation and tumor formation. TP53 mutation status serves as a key diagnostic and prognostic marker for many cancers, including breast, lung, and colorectal cancer. Emerging therapies are focusing on reactivating p53 function through gene therapy, small molecules, and immunotherapies, showing promising advancements in cancer treatment. Targeting the p53 protein for cancer therapy holds tremendous potential, and understanding its mutations and regulatory networks remains essential for developing novel cancer treatments.
