p53 Antibody (2B2.68) is an IgG2a kappa light chain mouse monoclonal antibody that binds to an N-terminal epitope between amino acid residues 11-25 of human p53, detecting both wild-type and mutant forms. p53 (2B2.68) monoclonal antibody is available for multiple applications such as immunoprecipitation, western blotting, immunohistochemistry, ELISA, flow cytometry, and immunofluorescence. The p53 protein, is a crucial tumor suppressor that plays a key role in maintaining DNA integrity. Encoded by the TP53 gene, p53 acts as a transcription factor, regulating essential cellular processes like DNA repair, cell cycle arrest, apoptosis, and cellular senescence. When cells experience stress or DNA damage, p53 activates and halts the cell cycle at critical checkpoints (G1/S or G2/M) to allow DNA repair. If the damage is irreparable, p53 initiates apoptosis, preventing the spread of genetically abnormal cells and reducing the risk of tumor development. p53 regulation occurs through post-translational modifications such as phosphorylation and acetylation. Phosphorylation at serine residues like Ser6, Ser15, and Ser46, by kinases such as ATM, ATR, and Chk2, enhances p53′s stability and ability to bind DNA. Acetylation by p300 and CBP stabilizes p53 further, while deacetylation by proteins like SIRT1 adjusts its function during cellular aging or in response to DNA damage. In cancer research, antibodies targeting p53 help study its role in disease. By using the p53 monoclonal antibody (2B2.68), researchers can assess p53 expression, post-translational modifications, and its localization in tissues. Given that p53 mutations are common in various cancers, these studies are critical for understanding tumor suppression mechanisms and developing targeted therapeutic strategies in the p53 pathway.

When should the TransCruz reagent sc-71817 X be used instead of the standard form sc-71817?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
