NMD3 encodes an essential ribosome biogenesis factor that functions as an adaptor for the export of pre-60S ribosomal subunits from the nucleus to the cytoplasm. By coordinating with the CRM1/XPO1 export pathway and remodeling events during late 60S maturation, NMD3 helps ensure productive assembly of translationally competent ribosomes and maintenance of proteostasis. Disruption of NMD3-dependent ribosome export can trigger nucleolar stress responses and alter cell-cycle progression through downstream effects on global translation. Because ribosome biogenesis is tightly linked to growth control, aberrant regulation of this process is relevant to studies of proliferative and stress-associated disease mechanisms, including ribosomopathies and cancer-associated translational rewiring.
NMD3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the NMD3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the NMD3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the NMD3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish NMD3 protein expression.
This CRISPR knockout system enables efficient generation of NMD3-deficient cell models for investigation of NMD3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
