MRTF-A Antibody (G-7) is an IgM κ mouse monoclonal MRTF-A antibody (also designated MKL1 antibody, MRTFA antibody, or myocardial related transcription factor A antibody) that detects the MRTF-A protein of mouse, rat and human origin by WB, IP, IF and ELISA. MRTF-A Antibody (G-7) is available as both the non-conjugated anti-MRTF-A antibody, and multiple Alexa Fluor® conjugates. Serum response factor (SRF) is a transcription factor that binds the serum response element (SRE), a sequence that mediates the transient response of many cellular genes to growth stimulation. SRF regulates the transient response of several muscle genes in response to growth factors and recruits accessory myogenic factors to activate these muscle genes. SRF is required for the formation of vertebrate mesoderm leading to the origin of the cardiovascular system. Myocardin, in association with SRF in cardiac muscle cells, activates cardiac muscle promoters. Myocardin-related transcription factor A (MRTF-A), also known as MKL1, interacts with SRF and stimulates its transcriptional activity.

Do you recommend G-7 or G-8 for IHC from 4% PFA fixed paraffin embedded tissue on 5 micron sections?

Thank you for your question. I would suggest sc-398675 for IHC use, although either one should be perfectly fine.

What secondary antibody do you suggest to use? Your data sheet says it's IgM light chain specific, all mouse IgM HRP I could find are specific for the heavy chain. However your data sheet recommends mouse IgG light chain specific HRP--very confusing......

Thank you for your question. We recommend using our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to Horseradish Peroxidase (HRP), m-IgGκ BP-HRP: sc-516102 (see link below). We do not offer a secondary binding protein that is specific to mouse IgM. However, our in-house data indicates that this anti-mouse IgG kappa light chain binding protein will also bind to the kappa light chain of mouse IgM. You could also use any conventional anti-mouse IgM secondary antibody. We do not offer any anti-mouse IgM that is specific for the heavy chain, so I'm not sure what you are referring to, unless those are provided by other suppliers. https://www.scbt.com/scbt/product/m-igg-kappa-bp-hrp

Can you recommend a positive control to use with MRTF-A (G-7): sc-398675 monoclonal antibody for Western blot?

We recommend Hep G2 (sc-2227), HEK293 (sc-45136), HUV-EC-C (sc-364180) or CCRF-CEM (sc-2225) whole cell lysates or HeLa nuclear extract (sc-2120) to use as positive controls as we have Western blot data showing specific detection of MRTF-A in these cell lines.

Rated 3 de 5 de par Unsure of specificityIt stains in the correct cellular area but its cellular localization is unspecific. I stained a fibrotic Human and Mouse Skin, liver and Kidney tissue. Staining is specific. However, I did not do an in vitro actin assay to truly determine antibody specificity. MRTF-A is published to be susceptible to actin destabilizing drugs.
