MRGX2 (also referred to as MRGPRX2 family member) encodes a Mas-related G protein–coupled receptor that participates in neuroimmune communication and stimulus-dependent signaling. In rodents, MRGX2-like receptors are linked to sensory neuron and mast cell–associated processes, coupling to downstream pathways that regulate intracellular calcium mobilization, degranulation programs, and inflammatory mediator release. As a GPCR, MRGX2 influences cell-surface signal transduction networks that intersect with stress and pain-related neurogenic inflammation, making it relevant for mechanistic studies of itch, nociception, and pseudo-allergic responses. Dysregulated activity in this receptor family has been associated with altered mast cell reactivity and inflammatory phenotypes in preclinical models.
MRGX2 CRISPR/Cas9 KO Plasmid (r) is a pool of plasmids designed for targeted disruption of the gene in rat cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the open reading frame following Cas9-mediated double-strand break formation.
