L'anticorps MEK-1/2 (9G3) est un anticorps monoclonal IgG2a de souris contre la protéine MEK-1/2 (également appelée anticorps MAP2K1, anticorps MAP2K2 ou anticorps contre la protéine kinase activée par un agent mitogène) qui détecte la protéine MEK-1/2 d'origine murine, rat, humaine et canine par WB et IP. L'anticorps MEK-1/2 (9G3) est disponible sous forme d'anticorps anti-MEK-1/2 non conjugué. Une famille de protéines kinases situées en amont des MAP kinases et responsables de leur activation a été identifiée. Le membre prototype de cette famille, appelé MAP kinase kinase, ou MEK-1, phosphoryle spécifiquement les résidus thréonine et tyrosine régulateurs des MAP kinases présents dans le motif Thr-Glu-Tyr d'ERK. Un deuxième membre de la famille MEK, MEK-2, ressemble à MEK-1 dans sa spécificité de substrat. MEK-3 (ou MKK-3) a pour fonction d'activer la MAP kinase p38, et MEK-4 (également appelée SEK1 ou MKK-4) active à la fois les MAP kinases p38 et JNK. La MEK-5 semble phosphoryler spécifiquement ERK5, tandis que la MEK-6 phosphoryle p38 et p38b. MEK-7 (ou MKK-7) phosphoryle et active la voie de transduction du signal JNK.

Rated 2 de 5 de par please use this antibody with caution!I have previously recommended this antibody. I have thought this antibody can equally recognise the phosphor- and non-phosphor-form of MEK-1/2, according to the statement "recommended for detection of the activation loop of MEK-1 and MEK-2 (independent of its phosphorylation status)". However, recently I did a growth factor stimulation experiment. I suddenly realised that MEK-1/2 Antibody (9G3) appears to have a strong preference for the p-MEK-1/2 over non-p-MEK-1/2, but is not totally clean with non-p-MEK-1/2. Thus, it would be problematic to use this antibody, especially when the phosphorylation level is low. So, please use this antibody with caution!

Rated 5 de 5 de par great for WBThis ab has very good specificity and sensitivity for WB with MEK-1/2 from human, mouse and rat. The epitope is in the activation loop, for sequential WB using a phosphor-MEK-1/2 and this Ab, whether the binding of one ab will interfere with the binding of the other is not unknown.
