LAIR-1 Antibody (NKTA255) is a mouse monoclonal IgG1 antibody that detects leukocyte-associated Ig-like receptor-1 (LAIR-1) in human samples through western blotting (WB), immunoprecipitation (IP), and flow cytometry (FCM). LAIR-1 is a transmembrane glycoprotein predominantly expressed on human peripheral blood mononuclear leukocytes, playing a crucial role in immune regulation. LAIR-1 functions as an inhibitory receptor on the cell surface, modulating immune responses by recruiting SHP-1, a phosphatase that negatively regulates cell signaling pathways. This interaction maintains the balance of immune activation and inhibition, particularly in T lymphocytes, where LAIR-1 helps establish signaling thresholds essential for T cell receptor (TCR) signaling. Phosphorylation of LAIR-1 at specific tyrosine residues, such as Tyr 233 and Tyr 263, facilitates SHP-1 recruitment and influences downstream signaling events. LAIR-1 engagement on myelomonocytic leukemic cell lines inhibits cell proliferation and induces programmed cell death, while LAIR-1 cross-linking on natural killer (NK) cells significantly dampens cytotoxic activity.
