HHV-8 Antibody (LN53) is a rat monoclonal IgG2c antibody that detects HHV-8 protein by immunofluorescence (IF) and immunohistochemistry with paraffin-embedded sections (IHCP). HHV-8, also known as Kaposi′s sarcoma-associated herpesvirus, plays a critical role in several malignancies, including multicentric Castleman′s disease and primary effusion lymphoma, which is a rare form of non-Hodgkin lymphoma that affects body cavities. HHV-8 K14 protein is particularly significant as K14 protein is expressed on the surfaces of infected cells exclusively during the lytic cycle, facilitating interaction with human CD200R, a receptor found on myeloid cells that helps regulate macrophage activation. This interaction is crucial because HHV-8 K14 protein inhibits tumor necrosis factor-alpha (TNFα) production, thereby locally restraining macrophage activation and contributing to viral evasion of host immune response. Additionally, HHV-8 encodes a viral cyclin homologous to cellular D-type cyclins, which are essential positive mediators of the cell cycle. Unlike cellular counterparts, HHV-8 cyclins are not regulated by p27 cell cycle inhibitor, which may elucidate the coexistence of p27 and high proliferative index observed in individuals with primary effusion lymphoma. Anti-HHV-8 antibody (LN53) is available as a non-conjugated format, making HHV-8 (LN53) antibody a valuable tool for researchers studying the pathogenesis of HHV-8-related diseases.

Can sc-73588: HHV-8 (LN53) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: DefinitelyNotMatt
Thank you for your question. Yes, sc-73588: HHV-8 (LN53) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
