GP-39 Antibody (E-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects GP-39 of mouse origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). GP-39 (E-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Human cartilage glycoprotein 39 (GP-39), also known as YKL-40, is a glycoprotein secreted primarily by articular chondrocytes, synoviocytes, and macrophages, playing a crucial role in the inflammatory response. Elevated levels of GP-39 in serum and synovial fluid are associated with inflammatory diseases, particularly rheumatoid arthritis, where GP-39 levels correlate with the extent of joint damage. GP-39 is predominantly expressed in articular chondrocytes and synovial cells, with additional expression in the liver, while GP-39 remains undetectable in muscle tissues, lung, pancreas, mononuclear cells, and fibroblasts. GP-39′s structure, characterized by a unique combination of a chitin-binding domain and a fibronectin type III-like domain, is essential for GP-39′s interaction with other proteins and role in modulating the immune response. GP-39 is considered a candidate autoantigen in rheumatoid arthritis, highlighting GP-39′s importance in the ability of cells to adapt to environmental changes and maintain homeostasis.

I am using GP-39 (E-11): sc-376910 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Professor Griffin
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
