Mouse Gata6 encodes the zinc finger transcription factor GATA-6, a key regulator of endodermal lineage specification and epithelial differentiation during embryogenesis and adult tissue homeostasis. GATA-6 modulates gene programs controlling cell fate decisions, morphogenesis, and organ development, including processes in the respiratory and gastrointestinal systems. It integrates with developmental transcriptional networks and signaling pathways such as WNT, BMP/TGF-β, and FGF to coordinate context-dependent transcriptional outputs. Dysregulated GATA-6 activity has been linked to developmental abnormalities and altered differentiation states that are relevant to modeling congenital defects and mechanisms of disease-associated tissue remodeling.
GATA-6 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Gata6 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Gata6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Gata6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish GATA-6 protein expression.
This CRISPR knockout system enables efficient generation of Gata6-deficient cell models for investigation of GATA-6 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
