Galectin-3 Antibody (M3/38.1.2.8 HL.2) is a rat monoclonal IgG2a antibody engineered to target galectin-3, also called Mac-2, hMac-2 (human Mac-2), GALBP (galactose-binding protein), CBP35 (carbohydrate-binding protein 35), and LGALS3 (lectin, galactoside-binding, soluble, 3). Anti-galectin-3 antibody (M3/38.1.2.8 HL.2) is raised against galectin-3 of mouse origin and is validated for the detection of galectin-3 in mouse, rat, and human samples using a variety of applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Galectin-3 is a pivotal member of the galectin family of soluble β-galactoside-binding lectins characterized by a single carbohydrate recognition domain (CRD) that is essential for binding to galactose-containing glycoconjugates. This structural feature allows galectin-3 to mediate critical biological processes such as cell-to-cell adhesion, interactions between cells and the extracellular matrix (ECM), and various cellular signaling pathways. Galectin-3 interacts with a range of specific proteins including β1 integrins, which are involved in cell adhesion and migration; the epidermal growth factor receptor (EGFR), which plays a key role in cell proliferation and survival; and members of the Bcl-2 family, which regulate apoptosis. Additionally, galectin-3 interacts with matrix metalloproteinases (MMPs) that are crucial for ECM remodeling and with RNA-binding proteins like heterogeneous nuclear ribonucleoproteins (hnRNPs) involved in pre-mRNA splicing. These interactions influence diverse cellular functions such as tumor progression, metastasis, apoptosis resistance, and immune responses, making galectin-3 a significant target in cancer research and other disease studies. Galectin-3 monoclonal antibody (M3/38.1.2.8 HL.2) is available in both non-conjugated form and conjugated forms including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatile options for diverse experimental applications.

Can galectin-3 (M3/38.1.2.8 HL.2): sc-81728 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: DefinitelyNotMatt
Thank you for your question. Yes, galectin-3 (M3/38.1.2.8 HL.2): sc-81728 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
