CTH Antibody (A-2) is a mouse monoclonal IgG2a antibody that detects CTH in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). CTH, or cystathionine γ-lyase, is a crucial enzyme in the trans-sulfuration pathway, primarily located in the cytoplasm, where CTH catalyzes the β-disulfide elimination reaction dependent on pyridoxal phosphate. This reaction produces ammonium, pyruvate, and thiocysteine, with the latter being a precursor for hydrogen sulfide (H2S) production. CTH′s cellular location is significant due to CTH′s role as the major H2S-producing enzyme in key tissues such as the kidney, liver, vascular smooth muscle cells, and enterocytes. The endogenous production of H2S is vital for regulating numerous cellular functions, including promoting cell growth, hyperpolarizing cell membranes, modulating neuronal excitability, and facilitating the relaxation of smooth muscle cells. Dysregulation or mutations in the CTH gene can lead to cystathioninuria, an autosomal recessive disorder characterized by the abnormal accumulation of cystathionine in plasma and increased urinary excretion, highlighting CTH′s importance in maintaining metabolic balance and cellular health. Anti-CTH antibody (A-2) is an essential tool for researchers investigating the physiological and pathological roles of this enzyme in various biological contexts.

I am using CTH (A-2): sc-365381 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Trav11
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
