Clusterin-α Antibody (A-11) is a mouse monoclonal IgG2a kappa light chain antibody engineered to recognize Clusterin-α of human origin by targeting an epitope mapped between amino acids 432-449 at the C-terminus. Clusterin-α monoclonal antibody (A-11) is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA), demonstrating robust reactivity with Clusterin-α from mouse, rat, and human species. Clusterin-α, also known as apolipoprotein J (ApoJ) or complement lysis inhibitor (CLI), is a multifunctional secretory heterodimeric glycoprotein composed of α and β subunits linked by disulfide bonds, formed through the post-translational cleavage of a 449 amino acid precursor at the internal bond between Arg227 and Ser228. The structural integrity of Clusterin-α is crucial for its diverse roles in various physiological processes, including immune regulation, cell adhesion, lipid transportation, tissue remodeling, membrane recycling, and cell-cell interactions. Functionally, Clusterin-α acts as a key modulator of the complement system by inhibiting the formation of the membrane attack complex, thereby protecting cells from complement-mediated lysis and maintaining immune homeostasis. Additionally, Clusterin-α interacts with apoptotic proteins such as Bax, regulating apoptosis and promoting cell survival under stress conditions. In the nervous system, Clusterin-α binds to amyloid-beta peptides, influencing their aggregation and reducing neuritic toxicity, which is significant in the pathogenesis of Alzheimer′s disease. Moreover, Clusterin-α interacts with various extracellular matrix proteins, facilitating cell adhesion and migration, which are essential for tissue remodeling and cancer metastasis. Overexpression of Clusterin-α has been observed in advanced stages of several cancers, including mammary tumors, where Clusterin-α supports tumor progression and metastasis by enhancing cell survival and resistance to apoptosis. Anti-Clusterin-α antibody (A-11) enables detailed studies of Clusterin-α′s interactions with these critical proteins, providing valuable insights into its role in immune regulation, apoptosis, neuroprotection, and cancer biology.
