Can this antibody be used for human cell line western blotting experiment?

Thank you for your question. Anti-liver Arginase 1/ARG1 Antibody (E-2): sc-271430 is recommended for detection of Arginase 1 of mouse and rat origin by WB, IP, IF, IHC(P) and ELISA, not recommended for human cell line western blotting experiment.

I performed the western blotting using arginase I antibody (E-2). But M1 macrophage showed two band between 35~50 kDa and M2 macrophage showed one band between 35~50 kDa. Question: In case of two band, all two band are arginase 1? or isoform can be?

Does this antibody specifically recognize Arg-1, or is there some cross reactivity with Arginase-2?

Thank you for your question. This mouse monoclonal antibody is raised against an antigen that is specific for mouse and rat arginase I, and does not cross react with human. The antigen is specific for arginase I, with no significant similarity to arginase II.

I got a beautiful band at the correct size on liver samples but I got a higher band for serum samples (50 kDa) no band at 38 kDa. Do you know what is this band?

Thank you for your question. We have not tested this mouse monoclonal, arginase I Antibody (E-2): sc-271430, on serum samples, nor have we observed band ~ 50 kDa when using the antibody for Western blotting. We would recommend including a secondary antibody control (omit primary antibody) to determine if the 50 kDa band may be a result of non-specific binding of your secondary antibody alone. A non-specific band ~ 50 kDa in serum samples could result from binding to the heavy chain of IgG.

Can arginase I Antibody (E-2): sc-271430 be used to stain paraformaldehyde-fixed, frozen tissue sections embedded in cryomatrix?

Thank you for your question. Yes, this mouse monoclonal, arginase I Antibody (E-2): sc-271430, is suitable for this research application. There is a recent publication that cites the antibody for PFA-fixed frozen tissue sections, reference, PMID: 25889790. If fixing in formalin, we recommend you include a cell permeabilization treatment. For example, incubating for 3 minutes in a solution of 0.01% Triton X-100 in PBS.

If the level of expression is very high, the Ab works perfectly. If the Arginase1 level is low, signal could be missed as there is some background present.

Rated 2 de 5 de par Western blot analysis NOS II and Arg-1 were detected using the appropriate antibodies at a dilution of 1:750 and 1:1000, respectively (Santa Cruz Biotechnology, CA, USA), with overnight incubation at 4 °C. Goat anti-rabbit peroxidase-conjugated antibody and goat anti-mouse peroxidase-conjugated antibody (Jackson ImmunoResearch; West Grove, PA, USA) were used at a 1:25,000 dilution as the secondary antibody. Membranes were stripped and incubated with anti-β-tubulin antibody (Santa Cruz Biotechnology CA, USA) at a dilution of 1:2,500 as a loading control.
