BKS Activators are a select group of chemical compounds that facilitate the enhancement of BKS activity through their influence on various intracellular signaling pathways. Forskolin and IBMX both lead to a rise in intracellular cAMP levels, resulting in the activation of PKA, a kinase that could phosphorylate and thereby enhance the activity of BKS. Similarly, EGCG, acting as a kinase inhibitor, and Sildenafil, through elevation of cGMP levels and subsequent PKG activation, could augment BKS activity. This is in addition to A23187 and PMA, which, by increasing intracellular calcium levels and activating PKC respectively, potentially promote the phosphorylation and activation of BKS. LY294002 and U0126 both function as inhibitors of the PI3K and MEK1/2 enzymes, respectively, and could shift cellular signaling dynamics in a manner that indirectly supports BKS pathway activation.

Furthermore, SB203580 and PD98059, as specific inhibitors of p38 MAPK and MEK, might create an intracellular environment that favors the activation of BKS by reducing competitive signaling. Thapsigargin, an inhibitor of SERCA, results in an increase in cytosolic calcium, which could activate calcium-dependent proteins that in turn activate BKS. Lastly, Spermine, by modulating ion channels and possibly interacting with calcium signaling, could also facilitate an increase in BKS activity. The combined actions of these compounds, through distinct but converging pathways, serve to enhance the functional activity of BKS without directly affecting its expression levels or requiring direct binding to the protein itself, ensuring that BKS is more actively involved in its respective cellular functions.
