STRA6 encodes Stra6, a multi-pass membrane receptor that mediates cellular uptake of vitamin A by binding retinol–retinol binding protein (RBP) complexes at the cell surface. By controlling intracellular retinoid availability, Stra6 influences retinoic acid signaling and downstream transcriptional programs that regulate differentiation, epithelial homeostasis, and developmental patterning. STRA6 activity intersects with retinoid metabolism networks involving LRAT, RDH/ALDH enzymes, and nuclear retinoic acid receptors, thereby shaping cell fate decisions and tissue integrity. Dysregulation or genetic variation in STRA6 has been linked to developmental abnormalities and vision-related phenotypes consistent with perturbed retinoid transport and signaling.
Stra6 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the STRA6 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the STRA6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the STRA6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Stra6 protein expression.
