Periplakin Antibody (AE11) is a mouse monoclonal IgG1 antibody that detects periplakin in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Periplakin is a crucial member of the plakin family of proteins, primarily involved in the structural integrity of desmosomes and the epidermal cornified envelope, which are essential for maintaining cell adhesion and barrier function in epithelial tissues. Periplakin is localized to desmosomes, the interdesmosomal plasma membrane, and intermediate filaments, where anti-periplakin antibody (AE11) helps reveal its vital role in linking the cytoskeleton to cell junctions. Notably, the N-terminus of periplakin accumulates at the cell surface of microvilli, associating with cortical actin, which is important for cellular signaling and maintaining the architecture of the cell surface. The rod domain of periplakin is essential for the redistribution of envoplakin to desmosomes and the cytoskeleton through heterodimerization, while the linker domain is necessary for association with intermediate filaments.
