p-p53 Antibody (F-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects p53, specifically the Ser 46 phosphorylated form, in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). p-p53 monoclonal antibody (F-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. p53, a crucial tumor suppressor, plays a vital role in regulating cell cycle and promoting apoptosis in response to cellular stress, thereby acting as a guardian of genomic integrity. p53 is primarily located in the nucleus, where p53 binds to DNA and activates transcription of genes involved in growth arrest and apoptosis. Phosphorylation of p53 at Ser 46 enhances pro-apoptotic activity by selectively transactivating genes involved in programmed cell death, making this modification critical in cellular stress responses. This phosphorylation event predominantly occurs in the nucleus, where p53 orchestrates transcriptional programs, but can also influence subcellular localization and stability, further regulating tumor-suppressive functions. p53 can be sequestered in the cytoplasm by E3 ubiquitin ligase MDM2, which is a negative regulator that targets p53 for proteasomal degradation. Dynamic localization of p53 is essential for function, allowing rapid responses to cellular stress signals. p53 undergoes various post-translational modifications, including phosphorylation and acetylation, which are critical for activation and function. Mutations in the DNA-binding domain of p53 can lead to loss of function and are frequently associated with a wide range of human cancers, underscoring p53′s importance in tumor suppression and anti-p53 antibody (F-11)′s potential in cancer research and diagnostics.

I wonder why the band size of this antibody (p-p53 Antibody (F-11): sc-377561) is at 89KDa when the predicted molercular weight is 53KDa. I bought this antibody and I get a band at around 100KDa?

Asked by: HRC1
Thank you for your question. The expected molecular weight of p-p53, as detected by p-p53 Antibody (F-11): sc-377561, is 53 kDa. If you are detecting a signal at a different height, please contact our technical service for assistance.

What application is the blocking peptide sc-377561 P appropriate for?

Asked by: TinTin
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
