The proliferation of eukaryotic cells is controlled at specific points in the cell cycle, particularly at the G1 to S and the G2 to M transitions. It is well established that the Cdc2 p34-cyclin B protein kinase plays a critical role in the G2 to M transition while cyclin A associates with Cdk2 p33 and functions in S phase. Considerable effort directed towards the identification of G1 cyclins has led to the isolation of cyclin D, cyclin C and cyclin E. Of these, cyclin D corresponds to a putative human oncogene, designated PRAD1, which maps at the site of the Bcl-1 rearrangement in certain lymphomas and leukemias. Two additional human type D cyclins, as well as their mouse homologs, have been identified. Evidence has established that members of the cyclin D family function to regulate phosphorylation of the retinoblastoma gene product, thereby activating E2F transcription factors.

Can cyclin D2 (DCS-5): sc-53637 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, cyclin D2 (DCS-5): sc-53637 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Specific Western detection in rat INS-1 cells60 ug loads of rat insulinoma (INS-1) lysates were probed overnight and returned a strong, specific band of ~38 kD. Antibody sc-56305 is also suitable. The attached picture is a triplicate set (identical lanes)

Nice signal by WB on mouse colon extractsThis antibody works perfectly on mouse colon extracts (20 ug) at a 1/200 dilution with regular ECL.

a relative good antibody for WBThis antibody generates a signal at the predicted position on the WB (1:500 dilution, 40ug HeLa extract). However, I would recommend to use sc-376676 for WB.

not very cleanIn HeLa, it is a low-affinity antibody. Not very clean, it has many nonspecific larger MW bands which are stronger than the cyclin-D2 band.
