BMPER encodes the secreted extracellular matrix glycoprotein Crossveinless-2, a context-dependent regulator of bone morphogenetic protein (BMP) signaling. By binding BMP ligands and modulating their availability to BMP receptors, BMPER influences SMAD-dependent transcriptional programs that control endothelial behavior, tissue morphogenesis, and extracellular matrix remodeling. BMPER activity intersects with developmental and vascular homeostasis pathways and has been linked in the literature to dysregulated angiogenic signaling and connective tissue phenotypes when perturbed. These properties make BMPER a useful target for studying BMP gradient formation, ligand sequestration, and pathway tuning in cell and organoid models.
Crossveinless-2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the BMPER gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the BMPER together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the BMPER open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Crossveinless-2 protein expression.
This CRISPR knockout system enables efficient generation of BMPER-deficient cell models for investigation of Crossveinless-2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
