Col17a1 encodes collagen type XVII alpha 1 (COL17A1), a transmembrane collagen and core component of hemidesmosomes that anchors basal keratinocytes to the underlying basement membrane. COL17A1 interacts with laminin-332 and integrin complexes to support epidermal adhesion, basement membrane organization, and skin appendage homeostasis, and its regulated ectodomain shedding influences keratinocyte migration and wound-associated remodeling. In mouse tissues, COL17A1 contributes to epithelial integrity and mechanotransduction at the dermal–epidermal junction, linking extracellular matrix cues to cytoskeletal stability. Disruption of COL17A1 function is broadly relevant to studies of skin fragility, blistering phenotypes, hair follicle cycling, and inflammation-associated epithelial barrier defects.
COL17A1 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Col17a1 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Col17a1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Col17a1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish COL17A1 protein expression.
This CRISPR knockout system enables efficient generation of Col17a1-deficient cell models for investigation of COL17A1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
