CLDN18 encodes claudin-18, a tetraspan tight-junction protein that contributes to paracellular barrier function and epithelial polarity in tissue-specific contexts, including gastric and pulmonary epithelia. By organizing tight junction strands with other claudins and scaffold proteins such as ZO family members, claudin-18 helps regulate junctional integrity, ion selectivity, and signaling linked to cytoskeletal dynamics. Altered CLDN18 expression or junctional localization is associated with epithelial remodeling and dysregulated barrier homeostasis, processes often connected to inflammation and tumor biology. CLDN18 has also been studied in the context of fusion events and lineage programs that impact cell adhesion and differentiation states.
claudin-18 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the CLDN18 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the CLDN18 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the CLDN18 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish claudin-18 protein expression.
