CD4 Antibody (OX38) is a mouse monoclonal IgG2a kappa light chain antibody designed to recognize CD4 protein, also known as T4 or Leu 3, in both mouse and rat species. Raised against MLR-generated T cells of rat origin, anti-CD4 antibody (OX38) is recommended for use in immunofluorescence (IF), immunohistochemistry (IHC), and flow cytometry (FCM), ensuring reliable detection of CD4 cell surface antigen across various experimental platforms. CD4 is a membrane-bound glycoprotein composed of four extracellular immunoglobulin-like domains, which are essential for structural integrity and function. Post-translational modifications, particularly glycosylation, play a crucial role in maintaining CD4′s stability, proper cell surface localization, and interactions with other proteins, thereby influencing T cell activation and immune responses. Located on the cell membrane, CD4 acts as a co-receptor for T cell receptor (TCR), enhancing binding affinity between TCRs and class II major histocompatibility complex (MHC) molecules on antigen-presenting cells. This interaction is vital for effective T cell activation and initiation of adaptive immune responses. Additionally, CD4 serves as the primary receptor for human immunodeficiency virus (HIV), facilitating viral entry into host cells and contributing to immune system impairment in HIV-infected individuals. CD4 interacts with key intracellular proteins such as tyrosine kinase Lck, which is essential for downstream signaling pathways that drive T cell proliferation and differentiation. By targeting structured extracellular regions of CD4, CD4 (OX38) monoclonal antibody enables precise identification and characterization of CD4-expressing cells, supporting critical research in immunology, including studies on autoimmune diseases, infectious diseases, and development of immunotherapies.
