CD4 Antibody (H129.19) is a rat monoclonal IgG2a antibody engineered to recognize CD4, also known as T4 or Leu 3, specifically in mouse samples. Anti-CD4 antibody (H129.19) is raised against the A.TH. mouse CTL clone A15.1.17 epitope and is recommended for use in immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM) applications, ensuring precise detection of CD4 in various experimental contexts. CD4 is a membrane-bound glycoprotein composed of four extracellular immunoglobulin-like domains, which are crucial for its function as a co-receptor in T cell activation by enhancing the interaction between the T cell receptor (TCR) and class II major histocompatibility complex (MHC) molecules on antigen-presenting cells. Post-translational modifications, such as glycosylation, play a vital role in maintaining CD4′s stability, proper cell surface localization, and its interactions with other proteins, thereby influencing the effectiveness of the immune response. Additionally, CD4 serves as a primary receptor for the human immunodeficiency virus (HIV), making CD4 a significant target for studies on viral entry and immune system regulation. The strategic localization of CD4 on T helper cells, cortical cells, mature medullary thymocytes, microglial cells, and dendritic cells highlights CD4′s importance in both adaptive and innate immunity. CD4 monoclonal antibody (H129.19) enables accurate identification and characterization of CD4-positive cells, supporting essential research in immunology, including investigations into autoimmune diseases, infectious diseases, and the development of immunotherapies.

Does this antibody work in IHC of sections from paraffin-embedded samples? Thanks for your help, Staffan

Asked by: Staffan
Thank you for your question. For staining paraffin-embedded, formalin-fixed tissue sample sections we would rather recommend using one of the two following alternate CD4 antibodies. We have several pieces of data and product citations for these products in the entiond application IHC(P). CD4 (MT310): sc-19641 https://www.scbt.com/scbt/product/cd4-antibody-mt310 CD4 (GK1.5): sc-13573 https://www.scbt.com/scbt/product/cd4-antibody-gk1-5 We suggest using the directly -HRP conjugated primary antibody according to our protocols: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining if you have any futher questions, please contact our Technical Service by phone or email or by Live Chat.

How can CD4 (H129.19): sc-19642 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: TinTin
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-19642 PE) or fluoroscein (sc-19642 FITC).
