CD21 Antibody (A-3) is a mouse monoclonal IgG2b kappa light chain antibody that detects CD21 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-CD21 antibody (A-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD21, also known as complement receptor type 2, is a type I integral membrane glycoprotein that plays a crucial role in the immune system by serving as a receptor for the C3d complement fragment and the Epstein-Barr virus. CD21 expression on mature B cells, follicular dendritic cells, and certain epithelial cells is vital for B cell activation and proliferation, as CD21 enhances the signaling strength of the B cell antigen receptor (BCR) in conjunction with the co-receptor CD19. This interaction is essential for the adaptive immune response, as CD21 modulates the activation threshold of B cells, ensuring a robust and specific response to foreign antigens. Additionally, CD21 undergoes phosphorylation following B cell activation, which is a key post-translational modification that influences downstream signaling pathways and cellular outcomes. Understanding CD21 function and regulation is important for elucidating mechanisms of immune response and potential therapeutic targets in diseases such as autoimmune disorders and infections.

Can CD21 (A-3): sc-13135 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD21 (A-3): sc-13135 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 1 out of 5 by from IHC (P): only for follicular dendritic cellsFormol-fixed (24h) paraffin sections of normal human spleen, antigen retrieval tested: 1) autoclaving in citrate buffer pH 6.0, or 2) protease XIV, or 3) no pretreatment: Only follicular dendritic cells (FDCs) stained, CD21-positive B-lymphocytes in follicles remain almost unstained. Best retrieval: autoclaving at pH 6.0. It may be possible to also stain B-lymphocytes if tyramide amplification is used in a peroxidase system.
