Aqp8 encodes aquaporin 8 (AQP8), a membrane channel that facilitates rapid water movement and can also contribute to transmembrane transport of small neutral solutes such as hydrogen peroxide in selected cell types. In mouse tissues, AQP8 expression is associated with epithelial fluid handling and secretory physiology, and subcellular localization has been reported in both plasma membrane and intracellular membranes, supporting roles in osmotic balance and organelle homeostasis. By influencing water permeability and redox-related solute flux, AQP8 intersects with processes including epithelial barrier function, mitochondrial physiology, and oxidative stress signaling. Dysregulated AQP8 activity or expression has been investigated in the context of inflammatory and metabolic phenotypes, liver and gastrointestinal physiology, and broader perturbations of cellular redox homeostasis relevant to disease mechanisms.
Aquaporin 8/AQP8 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Aqp8 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Aqp8 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Aqp8 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Aquaporin 8/AQP8 protein expression.
This CRISPR knockout system enables efficient generation of Aqp8-deficient cell models for investigation of Aquaporin 8/AQP8 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
