PBP Inhibidores

Penicillin-Binding Protein (PBP) inhibitors are a class of antibacterial agents, primarily consisting of beta-lactam antibiotics such as penicillins, cephalosporins, monobactams, and carbapenems. These inhibitors are crucial in the involvement of bacterial infections due to their specific action on PBPs, which are essential enzymes for bacterial cell wall synthesis. The mechanism of action of PBP inhibitors involves the mimicking of the D-Ala-D-Ala dipeptide, which is a natural substrate of PBPs. These antibiotics bind to the active site of PBPs, thereby inhibiting the transpeptidase activity necessary for cross-linking peptidoglycan chains. This inhibition leads to a weakened cell wall and eventually causes bacterial lysis, particularly effective during bacterial cell division.

Penicillins, including Penicillin G, Amoxicillin, and Oxacillin, were among the first PBP inhibitors discovered and have been widely used for decades. They are effective against a range of Gram-positive bacteria, and some are modified to be resistant to penicillinase, an enzyme produced by certain bacteria to inactivate penicillin. Cephalosporins, such as Cefazolin, Cefuroxime, and Ceftriaxone, represent another major class of PBP inhibitors. They are structurally similar to penicillins but with a broader spectrum of activity, including efficacy against many Gram-negative bacteria. Each subsequent generation of cephalosporins has expanded the range of bacterial susceptibility. Carbapenems like Meropenem and Imipenem are known for their broad-spectrum activity and resistance to many beta-lactamases. Aztreonam, a monobactam, is unique for its exclusive activity against Gram-negative bacteria, including Pseudomonas aeruginosa.
