    UDP-GlcDH (auch als UDP-Glucose 6-Dehydrogenase, UGDH oder UDPGDH bezeichnet) ist ein Mitglied der UDP-Glucose/GDP-Mannose-Dehydrogenase-Familie. UDP-GlcDH konvertiert UDP-Glucose in UDP-Glucuronsäure, die ein wesentlicher Bestandteil der Biosynthese von Glycosaminoglykanen, Hyaluronan, Heparansulfat und Chondroitinsulfat ist. Diese Glycosaminoglykane, die als gemeinsame Komponenten der extrazellulären Matrix vorkommen, sind von Bedeutung für die Signaltransduktion, Zellmigration, Tumorwachstum und Metastasierung. UDP-Glucuronsäure (UDP-GlcA) wird in der Leber für die Ausscheidung toxischer Substanzen benötigt. UDP-GlcDH ist ein ubiquitär exprimiertes Protein, das am häufigsten in der Leber vorkommt. Die Proteinstruktur von UDP-GlcDH wurde zuerst in der Kuhleber analysiert und als Homohexamer gefunden. Diese Struktur ist zwischen Spezies und Phyla gut konserviert, wobei zwischen verschiedenen Arten von Säugetieren eine übergeordnete Sequenzidentität von 97% besteht. Forschungsergebnisse deuten darauf hin, dass die Expression von UDP-GlcDH durch TGFβ aufgeregt und durch Hypoxie herabgesetzt wird.

    What blotting conditions were used for the blot? dilution? blocking agent? I used the antibody at 1:1000 in 5% milk/TBST. It was almost clean enough but could be a little better.

    Thank you for your question. You can find dilution recommendations for each antibody on their respective datasheets. Our protocol for western blotting that is used by our labs as well can be found under this link: https://www.scbt.com/resources/protocols/western-immuno-blotting We recommend UltraCruz® Blocking Reagent: sc-516214. Alternatively, a milk-based blocking buffer will produce a cleaner blot while BSA will provide stronger signals. Incubating the blot at a low dilution over night at 4 °C can also help to produce a robust and specific signal in many cases.
