UBTF encodes upstream binding transcription factor (UBF), a high-mobility group (HMG) box DNA-binding protein that is essential for RNA polymerase I–dependent transcription of ribosomal DNA. UBF helps organize rDNA chromatin, promotes formation of the Pol I preinitiation complex, and supports nucleolar structure and ribosome biogenesis, thereby linking it to cellular growth control and proteostasis. Through its role in nucleolar stress signaling and coordination with cell-cycle and DNA damage response pathways, altered UBTF/UBF activity can influence proliferation and genome stability. Dysregulation of Pol I transcription and nucleolar function is a recurrent feature of cancer biology and has also been implicated in neurodevelopmental and neurodegenerative disease contexts.
UBF CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the UBTF gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the UBTF together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the UBTF open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish UBF protein expression.
This CRISPR knockout system enables efficient generation of UBTF-deficient cell models for investigation of UBF signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
