
SPA-1 Antibody (B-2) is a mouse monoclonal IgG1 antibody that detects SPA-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SPA-1, or signal-induced proliferation-associated gene-1, serves as a crucial RAP1 GTPase-activating protein primarily found in hematopoietic progenitors and peripheral T cells, playing a significant role in immune response regulation. SPA-1 is predominantly expressed in fetal and adult lymphohematopoietic tissues, and SPA-1 expression is notably upregulated in normal lymphocytes following various mitogenic stimuli. By disrupting LFA-1-ICAM1-mediated adhesive interactions, SPA-1 inhibits T cell receptor triggering and interleukin-2 production, potentially through RAP1 inhibition. Studies show that mice lacking SPA-1 exhibit age-dependent T cell immunodeficiency, which can progress to late-onset myeloproliferative disorders, closely resembling human chronic myeloid leukemia. Additionally, SPA-1 interacts directly with aquaporin-2 (AQP2), influencing AQP2 trafficking to the apical membrane, which is vital for maintaining water homeostasis in renal function. With multifaceted roles in immune regulation and cellular signaling, SPA-1 is a key protein for understanding T cell dynamics and potential therapeutic targets in hematological disorders.
