RIMS3 encodes Rab3-interacting molecule 3 (Rim3), a presynaptic scaffolding protein that participates in organizing the neurotransmitter release apparatus at active zones. Through interactions with Rab3 GTPases and other synaptic components, Rim3 contributes to vesicle docking/priming, calcium-triggered exocytosis, and maintenance of synaptic efficacy. RIMS3-associated mechanisms intersect with neuronal signaling pathways governing synaptic transmission and plasticity, processes frequently implicated in neurodevelopmental and neuropsychiatric phenotypes. Altered expression or variation in RIMS3 has been investigated in the context of nervous system dysfunction, making it relevant for studying molecular determinants of synapse organization and network activity.
Rim3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the RIMS3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the RIMS3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the RIMS3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Rim3 protein expression.
