Ppp1cc encodes the mouse protein phosphatase 1 gamma (PP1γ), a catalytic serine/threonine phosphatase that controls phosphorylation-dependent signaling by dephosphorylating diverse substrates in the nucleus and cytoplasm. PP1γ functions within multiprotein holoenzymes that direct specificity toward processes including cell-cycle progression, chromatin dynamics, RNA processing, and cytoskeletal remodeling. Through these pathways, PP1γ contributes to regulation of mitotic exit, DNA damage responses, and cellular differentiation programs. Dysregulated PP1-family signaling and altered phosphorylation homeostasis are linked to mechanisms relevant to proliferative disorders, genomic instability, and neurobiology, supporting its use in pathway-focused functional studies.
PP1γ CRISPR/Cas9 KO Plasmid (m2) is a pool of plasmids designed for targeted disruption of the Ppp1cc gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Ppp1cc together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Ppp1cc open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PP1γ protein expression.
This CRISPR knockout system enables efficient generation of Ppp1cc-deficient cell models for investigation of PP1γ signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
