p-Cdc2 p34 Antibody (pY15.44) is a mouse monoclonal IgG1 kappa light chain antibody that detects p-Cdc2 p34 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). anti-p-Cdc2 p34 antibody (pY15.44) is available as the non-conjugated format. p-Cdc2 p34 protein, also known as p-CDK1, plays a crucial role in regulating the cell cycle, particularly in the transition from G2 phase to M phase. p-Cdc2 p34 is a serine/threonine-specific protein kinase that is tightly regulated through interaction with B-type cyclins and reversible phosphorylation. Binding of Cyclin B to Cdc2 p34 is essential for activation, promoting phosphorylation of Cdc2 at three key regulatory sites: threonine 14, tyrosine 15, and threonine 161. In higher eukaryotes, phosphorylation of threonine 14 and tyrosine 15 serves as a negative regulatory mechanism, preventing premature entry into mitosis. Conversely, phosphorylation of threonine 161 by cyclin-dependent kinases is necessary for Cdc2 activation and progression through the cell cycle. Precise regulation of these phosphorylation events is vital for ensuring proper cell division and maintaining genomic stability, highlighting p-Cdc2 p34′s importance in cellular function and development.

I am using p-Cdc2 p34 (pY15.44): sc-136014 for immunofluorescence, what dilution of this antibody should I use?

Rated 1 out of 5 by from Had bad luck with IFHad received sample of this antibody. This sample did not work for IF. I fixed my cells according to the well described method that involves 4% PFA (we fix at 4 degrees C). I used it in a cocktail of 1:100 dilution with other antibody (also in 1:100 dilution). The other antibody in the cocktail and DAPI worked as fine. Our secondary antibodies are great, so the only thing that I am not sure of is this antibody.
