NFXL1 CRISPR/Cas9 KO Plasmid (h) | sc-407389 | 20 µg | $397.00 |
NFXL1 encodes a zinc finger–containing transcriptional regulator implicated in controlling gene expression programs linked to cellular differentiation and stress-responsive signaling. As a putative DNA-binding factor, NFXL1 is expected to influence transcriptional networks that shape epithelial and immune-related processes, including pathways governing inflammatory tone and barrier function. Human genetic and functional studies have associated NFXL1 with neurodevelopmental phenotypes, supporting its relevance for investigating gene regulation in the context of brain development and cognitive function. Dissecting NFXL1-dependent transcriptional outputs can help clarify downstream targets and pathway crosstalk that contribute to complex disease-associated expression states.
NFXL1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the NFXL1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the NFXL1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the NFXL1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish NFXL1 protein expression.
This CRISPR knockout system enables efficient generation of NFXL1-deficient cell models for investigation of NFXL1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
